ارتباط چندشکلی های تک نوکلئوتیدی ژن BRCA1 با بیماری ورم پستان در گاوهای هلشتاین

چکیده

با استفاده از برنامه های اصلاح ژنتیکی می توان مقاومت نسبت به بیماری ها را در حیوانات افزایش داد. درانتخاب ژنتیکی علیه بیماری ورم پستان در گاوهای شیری از چندشکلی ژن کاندیدا استفاده می شود. چندشکلی ژن کاندیدا برای یافتن ارتباط میان SNP با فنوتیپ حیوان و به ویژه استفاده از آن در برنامه انتخاب به کمک نشانگر است. بدین منظور وجود چندشکلی اگزون 9ژن BRCA1 در 60 نمونه از گاوهای هلشتاین ایرانی با استفاده از روش PCR-RFLP بررسی شد که سه آلل مختلف در این بررسی شناسایی شدند. چندشکلی در جایگاه 46126+ از ژن BRCA1 با جهش G-A آشکار شد. فراوانی های ژنوتیپی به ترتیب برای سه نوع ترکیب ژنی بانامگذاری KK، KL وLL معادل 27/0، 43/0 و30/0 و دو آللی K وL برابر 48/0 و52/0 بدست آمد. برای مطالعه ارتباط ژنوتیپ و رکوردهای حاصل از شمارش سلول های بدنی از رویه GLMو رگرسیون لجستیک استفاده شد. افراد دارای ژنوتیپ KK به طور معنی داری نسبت به دو ژنوتیپ دیگر میزان سلول های بدنی کمتری داشتند. نتایج نشان داد که جایگاه مورد نظر در ژن BRCA1 به عنوان یک ژن مؤثر در مقاومت به ورم پستان می تواند در استراتژی های انتخاب به کمک نشانگر مورد استفاده قرار گیرد. چنین به نظر می رسد استفاده از تعداد نمونه بیشتر و بررسی ژن های کاندیدای دیگر می تواند برای دست یافتن به آزمونی با دقت بالاتر برای شناسایی حیوانات حساس و مقاوم به ورم پستان مؤثر باشد.
