بررسی ژنومی ساختار جمعیتی و عدم تعادل پیوستگی در برخی از نژاد‌های گوسفند بومی ایران

چکیده

فن‌آوری‌های ژنومی مانند تعیین ژنوتیپ با کارایی بالا بر اساس آرایه‌های SNP، پتانسیل بالایی برای رمزگشایی معماری ژنتیکی صفات پیچیده دارد و اطلاعات زمینه‌ای در مورد ساختار ژنوم در حیوانات اهلی، از قبیل میزان عدم تعادل پیوستگی (LD) فراهم می‌کند. در این پژوهش، از آرایه ژنومیIllumina Ovine SNP50K BeadChip برای تخمین و مقایسه عدم تعادل پیوستگی، اندازه مؤثر جمعیت (Ne) و هتروزیگوسیتی در سه جمعیت گوسفند ایرانی شامل گوسفند افشاری (41 رأس)، مغانی (35 رأس) و قزل (35 رأس) استفاده شد. میانگین LD (اندازه‌گیری شده توسط r2) بین SNP‌های مجاور با میانگین فاصله 58، 56 و 56 کیلو باز برای همه کروموزوم‌ها به‌ترتیب 207/0±151/0 برای نژاد افشاری، 190/0±131/0 برای نژاد مغانی و 184/0±121/0 برای نژاد قزل بود. کروموزوم 2 در نژاد افشاری و قزل و 12 در نژاد مغانی بیشترین میانگین r2 را در کروموزوم‌های اتوزوم هر نژاد داشتند. نژاد قزل بالاترین مقدار تنوع ژنتیکی را بر اساس اندازه مؤثر جمعیت و هتروزیگوسیتی نشان داد، در حالی که کمترین مقدار تنوع ژنتیکی در نژاد افشاری مشاهده شد. با توجه به مقادیر پایین عدم تعادل پیوستگی در این بررسی، نتایج مشخص کرد برای به‌دست آمدن صحت 85 درصدی در بررسی‌های انتخاب ژنومی و مطالعات پویش پیوستگی ژنومی به آرایه‌های با تراکم نشانگری بالاتر از 50Kb نیاز خواهد بود.
