بررسی اثر افزودن آنتی‌اکسیدان هایپوتائورین بر کاهش میزان پراکسیداسیون لیپید و بهبود فراسنجه‌های حرکتی اسپرم خروس طی انجماد و یخ گشایی

چکیده

هدف از مطالعه حاضر بررسی اثر آنتی‌اکسیدان ‌هایپوتائورین در سطوح 25/0، 5/0، 75/0 و 1 میلی‌مولار در رقیق‌کننده لیک بر پایه لسیتین طی فرایند انجماد و یخ‌گشایی بر روی فراسنجه‌های حرکتی، درصد زنده‌مانی، یکپارچگی غشای پلاسمایی، میزان مالون‌دی‌آلدهید موجود در مایع‌منی و مورفولوژی اسپرم‌ها بود. بدین منظور نمونه‌های منی از 15 خروس به روش مالش پشتی-شکمی بافاصله‌ی دو بار در هفته جمع‌آوری شدند. پس از رقیق‌سازی و افزودن سطوح مختلفی از آنتی‌اکسیدان‌ها، نمونه‌ها در بالای بخار ازت منجمد شدند. پس از یخ‌گشایی، نمونه‌ها ازنظر پارامتر‌های عملکردی اسپرم مورد ارزیابی قرار گرفتند. نتایج آزمایش نشان داد که افزودن سطح 75/0 میلی‌مولار آنتی‌اکسیدان هایپوتائورین باعث افزایش معنی دار میزان تحرک پیش‌رونده، درصد تحرک کل، فراسنجه VCL، افزایش درصد زنده‌مانی و افزایش یکپارچگی غشای پلاسمایی نمونه‌ها نسبت به گروه کنترل شد و همچنین سطح 75/0 میلی‌مولارسبب کاهش معنی‌دار میزان مالون‌دی‌الدهید موجود در مایع منی شد و بالاترین میزان ازنظر این پارامترها در این سطح به دست آمد (P<0.05). نتایج حاصل نشان داد که افزودن سطح بهینه از هایپوتائورین می‌تواند نقش محافظتی در برابر پراکسیداسیون لیپید داشته باشد و سبب بهبود فراسنجه‌های اسپرم خروس هنگام انجماد و یخ‌گشایی شود و آسیب‌های ناشی از تنش اکسیداتیو را کاهش دهد
