آنالیز بیوانفورماتیک پروموتر ژنهای مؤثر در بافتهای شکمبه و طحال گاوهای گوشتی تغذیه شده با علوفه و غلات

چکیده

عملکرد شکمبه در تغذیه با علوفه و غلات با هم متفاوت می‌باشد. همچنین حیوانات تغذیه شده با غلات نسبت به حیوانات تغذیه شده با علوفه بیشتر در معرض خطر احتمال ابتلا به ناهنجاری‌های متابولیک و بیماری‌های عفونی هستند. در این مطالعه به منظور درک بهتر شبکه تنظیمی درگیر در طحال و شکمبه گاوهای تحت تأثیر دو جیره غذایی متفاوت علوفه و غلات، در ابتدا ژن‌های با بیان متفاوت با استفاده از تجزیه و تحلیل داده‌های RNA-seq شناسایی شدند. آنالیز پروموتر با استفاده با پایگاه بیوانفورماتیکی Genomatix انجام شد. افزون بر این، برای ترسیم شبکه‌های ژنی مرتبط با فاکتور‌های رونویسی شناسایی شده در شکمبه و طحال از نرم افزار Cytoscape استفاده شد. در ادامه، نتایج تجزیه و تحلیل پروموتری منجر به شناسایی 31 فاکتور رونویسی جدید احتمالی در مراحل تنظیمی عملکرد شکمبه و 10 فاکتور رونویسی در طحال در گاو‌های تغذیه شده با علوفه و غلات گردید. بر اساس نتایج آمده، هم در شکمبه و هم در طحال 10 ژن شناسایی شدند که بیشترین میزان بیان را دارا بودند. با توجه به تجزیه و تحلیل نتایج در پایگاه اطلاعاتی DAVID، فرایندهای: کاهش اکسیداسیون، تنظیم تکثیر سلولی، انتقال یون، توسعه اپیدیدیم و توسعه اکتودرم، معنی دار ارزیابی شدند. بطور کلی نتایج بدست آمده از این مطالعه بینش ارزشمندی جهت درک مکانسیم‌های مولکولی طحال و شکمبه تحت تأثیر دو رژیم غذایی متفاوت علوفه و غلات فراهم می‌کند.
